You will not be able to update the BIOS with an unsupported CPU. Personally I would buy/borrow a cheap Skylake celeron and flash the BIOS with that then return it, make sure the retailer allows returns. That or take it to an IT store and get them to flash it for you.

The Desk Mini does not have a socketed BIOS ROM so a new BIOS chip is out of the question.

I bought a MB h110m-ITX/ac and am waiting for the relase of i5 7500 Kaby lake, I should upgrade the MB bios for the new cpu but I do not have another cpu to do that.
I will be able to update the bios directly with the new CPU? If not, how should I act?
