I have a question, if I connect a hard drive to M2_1, will pcie1 enter x8 mode then? Does this only happen when we connect the M2_5 drive ?

This is difficult to answer, without knowing which board, You are talking about
Take the Z790 PG Lightning

You see the first slot is connected to the CPU, trhe second slot, to the chipset, but it only has 4 lanes.
The M.2 PCIE 5.0x4 is also connected to the CPU, but as You can see it doesn't share resources, otherwise it will be mentioned in the specs, under Slots and Storage.
one tip, x16 has 2 meanings
The length of the slot 9.5cm
The number of lanes provided
PCIE4 in this case is a x16 slot (length), but has only contacts for 4 lanes, which makes it a PCIE x16 slot running at 4
But it is all in the specs, on the website
