Can the 2nd deskmini m.2 slot (back of motherboard) be used to boot from an m.2 ssd?

Yes, SSD installed to the back of the A300 Deskmini can be used as bootable SSD.

Note that both M.2 sockets only support NVMe SSDs and you can boot from either. I use the one on the back of the motherboard first as it's much easier to add a second SSD later to the socket on the top. They do not support SATA SSDs.
