Let's look at the picture of that connector, and recall that it is a 3 Watt stereo amplifier output speaker connection:

This is a two channel, 3 Watt stereo amplifier output connection. 'R' means Right, the Right channel, 'L' means Left, the Left channel. Front just means these are the two main stereo outputs, not any of the surround sound channels. Since they are all for the Front speakers, I will not include Front anymore.

This connector is meant to be used with a pair of small speakers, that you might have next to a monitor, just for casual PC sound and background music. You are not required to use this connector, it is just an added feature on this board.

If you do not have powered speakers, or an amplifier or receiver to use with some small speakers, that will be connected to the outputs on the board's IO panel,  then you could use this connection to power those small speakers.

So we have R- and R+ (Right negative and Right positive), and L- and L+ (Left negative and Left positive.)

R- and R+ connect to the - and + terminals of the right channel speaker, and the L- and L+ connect to the - and + terminals of the left speaker.

The "Please connect the Chassis speaker to this header" is a mistake, ignore it.
